Effectiveness of Garcinia mangostana (Mangosteen) pericarp’s crude extract as an alternative Hematoxylin component in Hematoxylin & Eosin staining technique.

Abstract

The application of natural dye from the pericarp's crude extract of Garcinia mangostana (mangosteen) for staining liver tissue from an indigenous material is more abundant and it composed of Anthocyanin that is stable to light so that it can be stored in a long time and still be viable, therefore the objective of this study was to investigate the mangosteen concentration extract and its staining property on rat's liver tissue. Pericarp's of mangosteen were extracted from DOST (Department of Science and Technology) using 75 % ethanol. Researchers divided it into two groups, group A which is the extracted pericarps of mangosteen with Ferric chloride as mordant and the group B is extracted pericarps of mangosteen with copper sulfate as mordant. Mordant has been used to make the staining faster and more efficient. The liver of rats was processed for paraffin, embedding technique and sectioned at 2 µm thickness. The sections were stained with group A and group B at 25 minutes. The results showed by both group A and group B could not stain nucleus of rats liver tissue. The level of stain such as poor was not significant to the Hematoxylin & Eosin technique; therefore pericarp's crude extract of Garcinia mangostana (mangosteen) could not be used as an alternative stain for hematoxylin in Hematoxylin & Eosin staining technique.

Objectives

The study determined the effectiveness of Garcinia mangostana as a hematoxylin component as manifested in the Hematoxylin and Eosin staining.
